RESEARCH INTERESTS
Malaria drug resistance, molecular epidemiology, clinical trials.
Dr. Plowe is chief of the Malaria Section in the Center for Vaccine Development. His group studies the molecular mechanisms and molecular epidemiology of resistance to antimalarial drugs, particularly the antifolate drugs and chloroquine. Plasmodium falciparum resistant to these drugs is already widespread in much of South America and Asia, and chloroquine resistance is responsible for large increases in malaria morbidity and mortality in parts of Africa. Dr. Plowe's team has developed rapid molecular assays for parasite mutations that cause the parasite to be resistant to specific drugs. Validating molecular assays for drug resistant malaria, transferring these technologies to laboratories in malaria endemic areas, and applying the assays for malaria control have been a major focus of Dr. Plowe's program. In collaboration with colleagues at the University of Mali, Dr. Plowe has also developed a field research site in the Dogon Country of Mali in West Africa. This field station is the site of studies of drug resistance, the human immune response to malaria and the pathogenesis of severe malaria, all in preparation for malaria vaccine trials. His team conducted the first malaria vaccine trial in Mali in Bandiagara in 2003-2004 and began another in late 2004. The Malaria Section also has a research program in Blantyre, Malawi in Central Africa, where the focus is on drug resistant malaria and clinical trials of malaria drugs.
