Cell Line Authentication

Increasingly, verification of cell lines is required by funding bodies and journals prior to acceptance of research papers and grant proposals. The Biopolymer/Genomics Core Facility offers human cell line authentication and detection of intra-species cell line cross-contamination using short tandem repeat (STR) DNA profiling. Nine human specific STR loci and the Amelogenin gender identification locus are co-amplified using the StemElite™ ID System (Promega Corporation). These loci collectively provide a genetic profile with a random match probability of 1 in 2.92 × 109. Additionally, the StemElite™ ID System incorporates a sensitive marker that specifically detects the presence of contaminating mouse (Mus musculus) DNA. The StemElite™ ID System includes all the standard American Type Culture Collection (ATCC) markers for cell line authentication.

Investigators provide the facility with DNA, and the facility will perform the PCR reaction, detection, and analysis and provide the investigator with an allele table. If a pair of samples is provided for comparison, an interpretation of the result will be issued.

  • Extracted DNA - 10-1000 μl at a minimum of 10 ng/μl
