Cell Line Authentication

Increasingly, verification of cell lines is required by funding entities and professional journals prior to acceptance of research papers and grant proposals. The Biopolymer/Genomics Core Facility offers human cell line authentication and detection of cell line cross-contamination using short tandem repeat (STR).  STR profiling is a rapid, reproducible, and standardized PCR-based method for the authentication of human cell lines. DNA profiling includes all the standard American Type Culture Collection (ATCC) markers for cell line authentication.  The GenePrint® 10 System allows co-amplification and three-color detection of nine human loci, including the ASN-0002 loci (TH01, TPOX, vWA, Amelogenin, CSF1PO, D16S539, D7S820, D13S317 and D5S818) as well as D21S11. These loci collectively provide a genetic profile with a random match probability of 1 in 2.92 × 109. 

Investigators provide the facility with a cell pellet or DNA; our facility will perform the extraction if a cell pellet was provided, PCR reaction, detection, analysis and provide the investigator with an allele table. If a pair of samples is provided for comparison, an interpretation of the result will be issued.

Sample Requirements:

  • Extracted DNA - 10-1000 μl at a minimum of 10 ng/μl
  • Cell Pellet

Results:

  • STR Allele Report
  • Results in 3 business days

Cost:

  • $85 per sample for single reactions
  • $100.00 per sample for DNA extraction and detection
