Research Interests

My scientific interest has been to determine mechanisms underlying association of obesity, a chronic positive energy balance, and development of metabolic diseases, e.g. Type 2 Diabetes. I built a research program at UMB focused on understanding contributions of cytosolic lipid droplets (CLD) to cellular and systemic lipid homeostasis in both physiological and pathological conditions, with funding support from NIH, American Diabetes Association (ADA) and American Heart Association (AHA). Key questions addressed in my laboratory are the following: what gene products regulate CLD degradation and their tissue specificity; how and what links CLD in adipose, liver, and muscle with metabolic disease? My work has contributed significantly to understanding how perilipin protein family members regulate tissue specific CLD hydrolysis via each member’s ability to protect the CLD neutral lipid core from key lipolytic enzymes.  My work also helped develop a concept that ectopic CLD are an adaptive response to changes in lipid homeostasis, and not necessarily pathologic. Rather, defects in CLD regulation and metabolism can lead to disease. My lab learned to use perilipin proteins as effective discovery tools to study CLD function, and we developed important tools and methodologies for the field to study CLD biology and lipid metabolism in cell culture systems and tissues.

            As the Director of the Biological Mechanisms Core Laboratory (NIH Mid-Atlantic Nutrition Obesity Research Center (NORC)), I developed strong interactions with the entire NORC scientific community, and in particular members involved in studying the genetics of complex disorders in the Old Order Amish including T2D, insulin resistance obesity and cardiovascular diseases (CVD). An example of this is illustrated by a major contribution made to elucidate the molecular mechanisms underlying a novel 19 base pair deletion that disrupts the coding region of the LIPE gene in Amish subjects with dyslipidemia. LIPE encodes hormone sensitive lipase (HSL), a key lipolytic enzyme, recently published in New England Journal of Medicine (with two press releases), and reviewed in Cell Metabolism. The accelerating pace of genomic discovery among NORC users has increased the need for mechanistic studies to elucidate the physiological roles of novel genes in cells, tissues, and whole organisms. To achieve this goal, with the help of my Co-Director Dr. Zaghloul, we have developed three types of services: i) adipose tissue phenotyping and tissue/cell banking; ii) whole animal functional genomics (zebrafish genetic models) and iii) functional genomics in tissue culture cells (including over-expression and knockdown in cultured cells, analytical and metabolic assays, and confocal imaging). Combining my expertise in CLD biology, lipid metabolism, and molecular and imaging technologies with Dr. Zaghoul expertise in zebrafish system, we are now evaluating the impact of TM6SF2 variants in human triglyceride-rich lipoprotein secretion. Our on-going takes advantage of a unique ‘genetic experiment of nature’ which offers opportunities to gain molecular insights in human regulation of TRL secretion, a pathway associated with unmet medical needs.
